بررسی مولکولی و فایلوژنتیک خرچنگ Chiromantes boulengeri (Decapoda، Brachyura، Sesarmidae) از اروند رود

مطالعه حاضر در سال 1393 به منطور درک صحیح از خصوصیات ژن mtDNA خرچنگ Chiromantes boulengeri (Calman, 1920) در آب های جزر و مدی اروند رود در منطقه جزیره مینو (پل مینو شهر و منطقه ام العجاج) انجام گردید. بدین منظور در مهر ماه سال 1393 گونه مورد مطالعه از پهنه های جزر و مدی و نواحی کم عمق جزیره مینو جمع آوری گردید و در الکل %70 نگهداری شدند و در آزمایشگاه دانشگاه علوم و فنون دریایی خرمشهر مورد بررسی قرار گرفت. سپس به منظور آنالیزهای مولکولی جهت بررسی ارتباط فایلوژنتیک این گونه با گونه های نزدیک در بانک ژنی NCBI، DNA نمونه ها با استفاده از روش فنل- کلروفرم استخراج و قطعه ژنی میتوکندریایی 16S rRNA توسط PCR تکثیر و در نهایت توالی یابی شدند. نتایج شناسایی های ریخت شناسی همراه با بررسی فیلوژنی توالی های به دست آمده از نشانگر میتوکندریائی، حاکی از وجود گونه C. boulengeri در منطقه مورد مطالعه می باشد. با استناد بر بررسی پلی مورفیسم، بین گونه های C. boulengeri دو منطقه جهش رخ داده است که بیانگر سیر تکاملی و سازش گونه C. boulengeri جهت بقای بهتر در شرایط مختلف اکولوژیکی در منطقه مورد مطالعه می باشد.
